How to use the Colorimeter

We are going to proceed to the counting of the doses with the colorimeter. For that, we will place 10 ml of the extender solution in each bottle, and, in one of them, we will add plus 0,4ml of pure semen with a Micropipette,(so we must do 2×0,2ml).

We turn on the colorimeter (ON/OFF), place the bottle standard (the one that only has extender solution) and determine the value of reference (which will be 0,0 ) pressing the keyboard(ZERO).

Next, we take off the bottle standard and replace it for the other one (the one that has extender solution + semen), previously well mixed, and press the keyboard (READ/DIRECT) obtaining a value.

The value obtained on the colorimeter will be “crossed” with the volume of the ejaculate: a value will appear,(which is the interception of these 2 values on the scale),which must be divided by the desired concentration of the doses we want to prepare. This concentration is determined after observation of the Spermatozoid’s (SPZ) abnormal shapes (rolled up tails, cytoplasmic drops and destroyed acrossomas).According to this observation, we will decide how many SPZ will contain each dose(3;3.5;and 4 m. m. SPZ).

This observation consists in, taking off a drop from the semen + extender solution bottle, using the Micropipette, and put it on a slide to analyse at the microscope.

Thus, depending on much or little presence of abnormal forms on the ejaculate, we will determine the concentration with which we want to make the doses. If, for example, we will decide that the semen must be diluted at 3m m SPZ (which corresponds not only at an ejaculate with little or any abnormal form but also to an ejaculate with good mobility, vitality and little agglutination).

The value obtained in the scale will be divided by 3 and we will immediately know the possible number of doses to obtain with this ejaculate.
